Description

LaBEaze Real Time qPCR platform allows the simultaneous amplification and detection of nucleic acids within a closed reaction vessel. The method is based upon the incorporation of energy transfer-labeled primers into the amplification product. The unique 3D folding structure of the primer blocks the fluorescent signal generation via energy transfer. The fluorescent signal is produced only when the primer is incorporated into a PCR product. The increment of the fluorescence signal directly correlates with the accumulation of PCR product at each cycle. The folded primers have an extremely low fluorescence eliminating the need to purify the PCR product prior to quantification; therefore, PCR and fluorescent signal detection can occur in a single reaction vessel. Signal is measured either during the reaction (real-time) or after the last cycle of the reaction (endpoint). Multiple fluorescent dyes can be associated with different primers and are suitable for SNP detection for both single nucleotide and In/Del mutations.  Melting curve analysis can also be used as a confirmative data to identify the truly positive alleles and eliminate the fail positive that is caused by the primer dimer iGentbio Krase Mutation kit is designed to detect some of Kras mutations in exon 12 and 13. KRAS mutations is a frequent driver of acquired resistance to anti-EGFR therapy in colorectal cancers. KRAS mutation is predictive of a very poor response to Panitumumab (Vectibix®) and Cetuximab (Erbitux®) therapy in colorectal cancer. Currently, the most reliable way to predict whether a colorectal cancer patient will respond to one of the EGFR-inhibiting drugs is to test for certain “activating” mutations in the gene that encodes KRAS, which occurs in 30%–50% of colorectal cancers. Studies show patients whose tumors express the mutated version of the KRAS gene will not respond to Cetuximab or Panitumumab. This product can be used to test patient sample DNA from tissue sample or from whole blood cells. The result will genotype both wide type and mutant. This product can’t be used alone for diagnostic purpose. USE FOR RESEARCH ONLY
